Deoxyribonuclease I (abbreviated as DNase I) is a deoxyribonuclease I obtained through recombinant expression of Pichia coli strains. This enzyme can efficiently digest single-stranded or double-stranded DNA fragments.
The activity of DNase Ⅰ is highly dependent on Ca2 +It is at a certain level and can be Mg2 +Or Mn2 +Activate. At Mg2 +Under the existing conditions, DNase Ⅰ can randomly cut DNA fragments from any site. In Mn2 +Under the existing conditions, DNaseⅠIt can be cleaved at approximately the same sites of double-stranded DNA to form a flat-end DNA fragment or a sticky end containing 1-2 nucleotide protrusions. When the working pH range is between 7 and 8, DNase Ⅰ exhibits the best endonuclease activity and can be applied to remove single-stranded or double-stranded DNA fragments from various RNA samples.
