AcuGenix™ Universal DNA Fragmentation module is a core pretreatment reagent solution designed to simplify and accelerate the preparation of next-generation sequencing (NGS) libraries. This product integrates the first three key steps - controllable DNA fragmentation, terminal repair, and 3 '-deoxyadenosine (dA) tailing - into a single-tube reaction system, eliminating the need for intermediate purification steps and significantly reducing manual operation time.

1. Fragment trimming with an additional step A:
Three-step enzymatic reactions can be completed in a single tube, eliminating three rounds of purification operations and minimizing the sample loss of precious low-initial quantity samples (such as circulating free DNA and FFPE paraffin-embedded DNA) to the greatest extent. The total manual operation time can be controlled within 45 minutes.

2. Precise and controllable
The size of the fragment can be precisely regulated by the total amount of DNA input and the reaction duration. By default, a uniform 200-500 bp fragment distribution is generated, perfectly compatible with Illumina and BGI MGI sequencing platforms. Parameter customization is supported to achieve target fragment screening.

3. Efficient End Repair and dA Tailing
The integrated reaction system converts the irregular and protruding ends of nucleic acids into flat-ended, 5' phosphorylated substrates. Subsequently, the Klenow fragment (Klenow exo⁻) lacking 3'→5' exonuctase activity was used to complete the dA tailing, adding a single dA base to avoid self-connection of the linker and ensure the directional connection with the T-tail linker.

4. Wide adaptability and high sensitivity:
It has been verified that the applicable range of sample input is 1 pg to 1 μg, and it can handle genomic DNA, circulating cell-free DNA (cfDNA), FFPE derived DNA and PCR products. The distribution of fragments between batches and the stability of connection efficiency are excellent.

5. Ultra-high purity
After strict testing, no nuclease activity was detected and no residual host genomic DNA was found, ensuring the integrity of sequencing data and the repeatability of the experiment.
